Maintenance and functional coupling of ER contacts with the plasma membrane

The endoplasmic reticulum (ER) differentiates a peripheral membrane network of sheets, tubules, and ER contacts with the plasma membrane (PM). While ER tubules were suggested to store intracellular Ca2+, in cardiac myocytes Ca2+ release through ryanodine receptors (RyRs) is clustered in ER sheets at thousands of PM contacts and separate from Ca2+ uptake through Ca2+-ATPase pumps. We investigate the domain nature of local ER Ca2+ regulation and how it interfaces with protein synthesis in ER cisternae through translocon and alternative pathways in cardiac myocytes.
