Requirements for transporting a large cargo through the permeability barrier of nuclear pores
We propose to establish a model system for studying transport of very large cargoes through nuclear pores, starting from transport-incompetent mature ribosomal subunits (rSUs). We will generate nanobodies against these rSUs, fuse them to nuclear transport signals and thus engineer a series of adapters that recruit exportins or importins to specific sites on the rSUs' surfaces. We will then ask, which arrangement confers the most efficient passage of rSUs through authentic nuclear pores. We will also explore how ribosome-conducting barriers can be self-assembled from individual FG domains.
